Read anytime really, I recommend near the end of the DH run Usagi Yojimbo: Legends from Dark Horse collects odds and ends. These are volumes 34 and 35 of the series, and are ongoing from IDW. The only part missing here is that IDW is publishing the new “Usagi Yojimbo: Color Classics” series, the first volume collected as “Usagi Yojimbo: Origins”, is a new colour reprinting of the whole series by the new publisher. Fantagraphics has the ones they originally published, Dark Horse has the Mirage and Dark Horse runs I don't know the reason why they couldn't collect the first volume in the Saga trades, but I'd assume Fantagraphics have some sort of publishing rights to them (someone more knowledgeable can correct me) When a recurring character appears, you'll usually be given a bit of context for how Usagi knows them. The characters have full and rich histories, but rarely so central to the story that it can't be understood if you don't know it. The stories are pretty episodic and most volumes are fine as a starting point. Bunraku and Other Stories is the first volume of new stories from IDW, Origins is the first collection of the newly colourized material, which starts from the Mirage issues and I believe is planned to go through all of the Mirage/Dark Horse material. They are both collecting new issues of Usagi and reprinting old collections, now in colour. More recently, he moved publishers over to IDW. There are 9 numbered "Saga" volumes, plus "Legends" which collect Usagi stories that take place outside of the normal timeline (most famously Space Usagi). Usagi Yojimbo Saga is the line of larger collections also from Dark Horse that collect multiple volumes' worth of issues.
